Genetic manipulation is no longer the province of the specialized researcher. It is finding widespread application in all fields of medicine and biology. Nevertheless, application of these relatively new techniques to new areas of research is often fraught with unexpected problems and difficulties. Based on the Society for Applied Bacteriology''s Autumn 1989 Conference, this unique volume covers a wide and very up-to-date range of techniques used in genetic engineering. These include the isolation and analysis of DNA and RNA from cells and tissues, the selection and use of phage and plasmic vectors for cloning DNA, the cloning procedures, the production and screening of genomic libraries, the production and use of DNA probes, the polymerase chain reaction and the synthesis of ''designer'' genes. This volume contains many examples of the applications of the above and other techniques for genetic manipulation, to subjects as diverse as plant pathology, forensic science, bacterial taxonomy, cardiac research, diagnostic microbiology, food hygiene and sewage treatment.

Organization and Practice in Tuberculosis Bacteriology focuses on the organization and technical methods of tuberculosis bacteriology. This book discusses the mycobacteria and mycobacterial disease and role and scope of the tuberculosis laboratory in human and veterinary medicine and epidemiology. The organization of tuberculosis laboratory services; collection, preservation and transport of specimens; and examination of direct smears are also elaborated. This text likewise covers the decontamination of specimens, inoculation of culture media, and identification of species and variants of tubercle bacilli. Other topics include the drug sensitivity tests, animal inoculation and assay tests; problems faced in the therapy of tuberculosis, and lipid chromatography. This publication is suitable for biologists, medical practitioners, and clinicians concerned with tuberculosis bacteriology.

In recent years there have been many technical developments in immunology that have potential or actual applications in many other branches of the life sciences. These developments include a number of ultrasensitive systems for the detection and assay of a wide range of biological materials, including micro-organisms and their products, as well as techniques for isolating and purifying micro-organisms and other antigenic materials. Two developments in particular have greatly extended the scope and applicability of immunological methods. These are the production of monoclonal antibodies and the introduction of enzyme linked immunoassay as a simple, safe and economical alternative to radioimmunoassay. This book is based on information gathered at the Society for Applied Bacteriology Autumn Demonstration Meeting held at the South Bank Polytechnic, London on October 15th, 1986.
