This volume focuses on the biomedical aspects of inorganic polyphosphates, a family of unique bio-inorganic polymers.
In recent years, great advances have been made in understanding the development, metabolism, and physiological role of inorganic polyphosphates. These energy-rich polymers, which consist of long chains of phosphate units, are evolutionary old molecules. The acidocalcisomes, conserved organelles from bacteria to humans, as well as the mitochondria play a central role in polyphosphate production and storage. Polyphosphates have been assigned multiple functions, some of which are closely related to medically important processes, such as blood coagulation and fibrinolysis, energy metabolism, cell cycle regulation, apoptosis, chaperon function, microvascularization, stress response, neurodegeneration and aging. The development of bioinspired polyphosphate particles, in combination with suitable hydrogel-forming polymers enabled the development of new strategies in regenerative medicine, in particular for hard and soft tissue repair, but also in drug delivery and antimicrobial defense.

Lake Baikal is the oldest, deepest and most voluminous lake on Earth, comprising one fifth of the World’s unfrozen fresh water. It hosts the highest number of endemic animals recorded in any freshwater lake. Until recently it remained enigmatic why such a high diversity evolved in the isolated Lake Baikal. Focusing on the sponges (phylum Porifera) as an example, some answers are provided to fundamental questions on evolutionary forces. The characteristic feature of these animals is that they form their polymeric silicic acid skeleton enzymatically. This process is explored using modern molecular biological and cellular biological techniques to outline strategies to fabricate novel materials applicable in biomedicine and nanooptics.

Based on the assumption that invertebrates as well as vertebrates possess factors regulating hematopoiesis, response to infection or wounding, studies dealing with the evolution of immunity have focused on the isolation and characterization of putative cytokine-related molecules from invertebrates. Until recently, most of our knowledge of cytokine- and cytokine receptor-like molecules in invertebrates has relied on functional assays and similarities at the physicochemical level. As such, a phylogenetic relationship between invertebrate cytokine-like molecules and invertebrate counterparts could not be convincingly demonstrated. In the present book, recent studies demonstrating cytokine-like activities and related signaling pathways in invertebrates are critically reviewed, focusing on findings from molecular biology and taking advantage of the completion of the genome from the fly Drosophila and the worm Caenorhabditis elegans.

During evolution silica deposition has been used in Protozoa, Metazoa and in plants as skeletal elements. It appears that the mechanisms for the formation of biogenic silica have evolved independently in these three taxa. In Protozoa and plants biosilicification appears to be primarily driven by non-enzymatic processes and procedes on organic matrices. In contrast, in sponges (phylum Porifera) this process is mediated by enzymes; the initiation of this process is likewise dependent on organic matrices.
In this monograph the role of biosilica as stabilizing structures in different organisms is reviewed and their role for morphogenetic processes is outlined. It provides an up-to-date summary of the mechanisms by which polymeric biosilica is formed. The volume is intended for biologists, biochemists and molecular biologists, involved in the understanding of structure formation in living organisms and will also be very useful for scientists working in the field of applied Nanotechnology and Nanobiotechnology.

Sponges (phylum Porifera) are known to be very rich sources for bioactive compounds, mainly secondary metabolites. Main efforts are devoted to cell- and mariculture of sponges to assure a sustainable exploitation of bioactive compounds from biological starting material. These activities are flanked by improved technologies to cultivate bacteria and fungi which are associated with the sponges. It is the hope that by elucidating the strategies of interaction between microorganisms and their host (sponge), by modern cell and molecular biological methods, a more comprehensive cultivation of the symbiotic organisms will be possible. The next step in the transfer of knowledge to biotechnological applications is the isolation, characterization and structural determination of the bioactive compounds by sophisticated chemical approaches.
