Written for a scientific audience, it is also applicable to non-specialists as well.- Provides an unique overview of what we know about addiction, and how scientific knowledge can and should be applied in the societal, ethical, and political context- Applies the state-of-the-art research in fields such as Genomics, Neuroscience, Pharmacology, Social Policy and Ethics to addiction research- Includes a preface by Sir David King, Science Advisory to the UK Government and head of the Office of Science and Technology, and in introduction by Nora Volkow, director of the National Institute on Drug Abuse in the USA

This Handbook brings together and integrates comprehensively the core approaches to fear and anxiety. Its four sections: Animal models; neural systems; pharmacology; and clinical approaches, provide a range of perspectives that interact to produce new light on these important and sometimes dysfunctional emotions. Fear and anxiety are analyzed as patterns that have evolved on the basis of their adaptive functioning in response to threat. These patterns are stringently selected, providing a close fit with environmental situations and events; they are highly conservative across mammalian species, producing important similarities, along with some systematic differences, in their human expression in comparison to that of nonhuman mammals. These patterns are described, with attention to both adaptive and maladaptive components, and related to new understanding of neuroanatomic, neurotransmitter, and genetic mechanisms. Although chapters in the volume acknowledge important differences in views of fear and anxiety stemming from animal vs. human research, the emphasis of the volume is on a search for an integrated view that will facilitate the use of animal models of anxiety to predict drug response in people; on new technologies that will enable direct evaluation of biological mechanisms in anxiety disorders; and on strengthening the analysis of anxiety disorders as biological phenomena.- Integrates animal and human research on fear and anxiety- Presents emerging and developing fields of human anxiety research including imaging of anxiety disorders, the genetics of anxiety, the pharmacology of anxiolysis, recent developments in classification of anxiety disorders, linking these to animal work- Covers basic research on innate and conditioned responses to threat- Presents work from the major laboratories, on fear learning and extinction- Reviews research on an array of neurotransmitter and neuromodulator systems related to fear and anxiety- Compares models, and neural systems for learned versus unlearned responses to threat- Relates the findings to the study, diagnostics, and treatment of anxiety disorders, the major source of mental illness in modern society (26 % of Americans are affected by anxiety disorders!)

This book covers the entire field of research in the area of minor tranquillizers and its application to current clinical practice in the treatment of anxiety and insomnia. These drugs are principally the benzodiazepines and related drugs with a similar mechanism of action, such as zolpidem and zopiclone. The molecular mechanism of action of benzodiazepines is described, focussing on the interaction of these drugs with the different isoform of the GABAA^O receptor, and the consequences of this for brain function. Recent advances in this knowledge have provided a framework for defining the physiochemical nature of the interaction between such drugs and their receptor protein, and thus pave the way for the design of new anxiolytic and hypnotic drugs. The animal models available for evaluating the potential of such new therapeutic agents in the treatment of anxiety and insomnia are discussed. Furthermore, understanding of the physiological regulation of the GABAA receptor may provide insights into the aetiopathology of these diseases. The clinical use of benzodiazepines and related drugs in the treatment of anxiety, insomnia, epilepsy and as anaesthetics are explored. The advantages and limitations of such treatments are discussed, and the impact of drugs evaluated. A chapter is devoted to the issue of independence, the clinical pertinence of tolerence and dependence, and evaluates treatment options that may minimise the risk of dependence.

Foreword Barry Jacobs once memorably commented that the problem in understanding the role of serotonin in the brain was that it was implicated in virtually eve- thing but responsible for nothing [1]. Serotonin (5-HT) is defnitely implicated in the regulation of sleep but what does it do? Once seen as a sleep-promoting molecule, Jacob’s own work, lucidly described in this volume, radically changed our view. In fact, serotonin neurons are most active during waking; their - tivity diminishes with sleep onset and disappears during rapid eye movement (REM) sleep. Serotonin, therefore, is intimately linked to the sleep-wake cycle, but what is its role, what mechanisms are involved and what are the impli- tions for sleep medicine and psychiatry? Making progress with these diffcult questions requires a “translational” approach, which is a research paradigm where both laboratory and clinical neuroscience inform and guide each other with the ultimate aim of improving understanding and treatment of medical conditions. This volume is a superb example of translational research, where leading basic and clinical scientists - tegrate molecular, neuropharmacological and systems approaches to illuminate the reciprocal interactions of serotonin neurons and the mechanisms involved in sleep and circadian regulation. Serotonin is an ancient chemical mediator preserved through at least 500 m- lion years of evolution. In invertebrate animals such as mollusks and leeches, 5-HT cells are distributed throughout the nervous system among the various ganglia.
