Avian Influenza has become one of the biggest threats for human and animal health. The old paradigm was that the disease in waterfowl, poultry, pigs and man was caused by separate viruses that each stayed reasonably well within their own niche. The only danger to man was considered being infected by pigs, being the mixing vessel, where avian and human influenza viruses could come together and exchange genetic material to form new viruses that are potentially dangerous to man.This has dramatically proven wrong during the last decade, with huge outbreaks in the USA, Europe, and Asia. The H5N1 strain that caused human deaths in Hong Kong appeared to be transmitted directly from poultry to man. This initiated sudden awareness that pigs were not a necessary intermediate in the transmission chain. During the AI outbreaks in Italy, mutation of low-pathogenicity viruses into high-pathogenicity viruses in poultry appeared another new threat, and further evidence that the poultry sector had a wolf in sheep’s clothing. It put pressure on development of diagnostic methods that could be used in large monitoring programmes.In The Netherlands a human fatality, after increased reports of conjunctivitis during a H7N7 outbreak, signalled that different AI strains could be fatal to man. Also, the huge economic losses and difficulties in controlling the spread of the infection in densely populated poultry areas, problems with vaccination and lack of marker vaccines demonstrated that the current control policy must be improved. Years ago when we were asked to write a book on the present-day knowledge of the molecular biology of poliovirus, we did not expect that such an apparently simple task could involve so much time and effort. Our writing was hampered by the fact that both of us are full time "workers", so that this monograph is mainly a spare time expedience. The main attention of this book focuses on a detailed review of the molecular biology of poliovirus and especially on the advances of the last decade; medical and environmental aspects are only briefly mentioned. Observations from older studies are considered in view of more recent information. Some of the older ob­ servations provided fundamental insights and paved the way for present day research; too often such data has been neglected or independently rediscovered. Today, poliovirus research has again attracted considerable interest. High points gained within the last few years were the elucidation of the complete nucleotide sequences of the RNAs of the three poliovinls serotypes and the corresponding vaccine strains, the demonstration of genome evolution during transmission of poliovirus in an epidemic, further characterization of the antigenic sites on the virus particle and of the antigenic drift, characterization of alternate conforma­ tional states of the virion capsid, the development of monoclonal antibodies against some of the virus proteins, observations on the role of the plasma membrane, cytoskeleton, and cytoplasmic membranes as mediators in the virus­ induced redirection of the synthetic machinery of the host cell, and characteriza­

This volume collects the contributions presented at the "Working Conference on System Theory in Immunology", held in Rome, May 1978. The aim of the Conference was to bring together immunologists on one side and experts in system theory and applied mathematics on the other, in order to identify problems of common interest and to establish a network of joint effort toward their solution. The methodologies of system theory for processing experimental data and for describing dynamical phenomena could indeed contribute significantly to the under­ standing of basic immunological facts. Conversely, the complexity of experimental results and of interpretative models should stimulate mathematicians to formulate new problems and to design appropriate procedures of analysis. The multitude of scientific publications in theoretical biology, appeared in recent years, confirms this trend and calls for extensive interaction between mat- matics and immunology. The material of this volume is divided into five sections, along the scheme of the Conference program.
